BD Accuri C6 Flow Cytometer

             BD Accuri C6 Flow Cytometer is afforadable, bench top and very user friendly flow cytometer analyzer from BD Biosciences. This flow cytometer is very useful for the research projects which involves analysis of regular fluorescence which are in the range of FITC, PE, PerCP and APC. The instrument has fixed voltage which makes it easy to load sample tubes and analyze. Sample tubes can be anything such as 0.5mL tubes, 1.5mL tubes, 5mL tubes, 24 well plate, 48 well plate, even 96 well plate can be used. Before and after analysis, the cytometer runs the cleaning and decontamination automatically.

Instrument is also installed with CSampler which makes it easy to load the tubes and walk way.



Lasers:
2 lasers (488nm Blue Laser and 640nm Red Laser)


Detectors: 530/30, 585/40, 670LP and 670/30.. (There are also optional detectors available from BD)

Software: Accuri C6 Software and CSample Software.

Accuri C6 software can acquire up to 10,000 events per second and data is digitally collected over a 7-decade dynamic range. The fluidics system in accuri can determine the sample volume and can calculate absolute counts. It can also detect the sample concentration per micro liter.

Protocol: Sample preparation for Cell Cycle Analysis by Flow Cytometry

Protocol:  Preparation of Samples for Cell Cycle Analysis by Flow Cytometry: 1) Suspend approximately million cells (10^6) in 1mL of phosphate-buffered saline solution (1x). And with the help of pipette, gently aspirate about 7-8 times, to make the single cell suspension.  2) Take a centrifuge tube and add 4mL of 70% ethanol. (70% ethanol should be one ice before using). and add 1mL of cells in PBS to this tube. Keep the tubes on ice for minimum 2 hours at 4℃.. This step will fix the cells.  ( cells can kept at 4 ℃ for a week).  3) Centrifuge the ethanol fixed cells for 5 minutes at 300g. Carefully take out the supernatant. Do not disturb the pellet.  4) Add 5mL of cold 1x PBS to this pellet and suspend the cells using pipette.  5) Centrifuge the tubes at 300g for 5 minutes. and discard the supernatant.
